Summary:Hibiscus sabdariffa L. also known as roselle, is widely used in food and beverage industries. However, climate change had disrupted the life cycle of roselle. Alternative propagation method in roselle should be widely studied to ensure the longetivity of roselle in industries. In this paper, a protocol was conducted for protoplast isolation from roselle explants using enzymatic approach. The main objective of protoplasts isolation is to detach single protoplast from the strong protection of cell wall with the stable cells arrangement in explants. A protoplast yield of 4.26×107 was obtained when 1 g of explant strips being soaked in enzyme solution contain of 0.7 M osmoticum with 1.5% cellulase and 0.025% macerozyme. The leaf strips had been incubated in the enzyme solution for 5–13 hours. The best incubation time recorded was at 7 hour. © 2017 American Scientific Publishers All rights reserved.
